Isolation and Molecular characterization of ¥ÕFC1, a New Temperate Phage from Entercoccus faecalis

Abstract
A new temperate phage was isolated from an Enterococcus faecalis strain, KBL 703. Ultraviolet light treatment of this strain resulted in the induction of the temperate phage (named 0FC1) in about 2 h after the intracellular replication. The morphology of 0FC1 was examined by electron microscope. This 0FC1 bacteriophage had an icosahedral head (d = 62 nm) and a sheathless non-contractile tail 0 = 75 nm). Therefore, i0FC1 could he classified into Bradley¢¥s group B and styloviridae in ICTV classification. Analysis of the structural proteins of this phage by sodium dodecylsulfate-polyacrylamide gel electrophoresis (SDS-PAGE) revealed more than 20 bands, most of which represent molecular weights of less than 40 kDa. Restriction endonuclease analysis of the nucleic acid from this 0FC1 particle showed that it had about a 40.5 kb double stranded DNA genome. A part of the genomic DNA from this phage was sequenced. Southern hybridization analysis showed that this phage DNA was incorporated in the host chromosomal DNA as a prophage.
