The Characteristic Binding of Mutant Cyclic AMP Receptor Protein (CRP*) to Lactose Promoter in Escherichia coli

Abstract
This study was conducted to characterize the binding of two mutant cAMP receptor proteins (CRP*s), 91 CRP and 222 CRP, to lactose promoter. Gel shift assay suggested that 222 CRP had a similar conformation like wild-type CRP-CAMP but 91 CRP did not. 91 CRP-cGMP complex formed the characteristic complex with lactose promoter whose ternary complex migrated very closely to the uncomplexed DNA. Ring closure experiment confirmed that 91 CRP-cGMP induced DNA bending with a higher bending angle.
