Transforming Grwoth Factor-¥â1 Derived from Murine Peyer¢¥s Patch T Lymphocytes Specifically Increases IgA Isotype Synthesis

Abstract
Purified transforming growth factor-01 (TGF-01) has been reported to act as an immunoglobulin A (IgA) isotype switching factor in vitro. The aim of this study was to find whether TGFP1 can be produced by intestinal Peyer¢¥s patch (PP) T cells and whether synthesized TGF-01 has such activity. Murine autoreactive PP T cell fine was stimulated with Concanavalin A and supernatants (Sn) were added to surface IgA negative (sIgA-) B cell culture. The acid-activated T cell Sn in the presence of Interleukin-2 induced a 10-fold or greater increase in IgA isotype production but not in IgM or IgGI isotypes by sIgA- B cells detected by ELISA. Furthermore, the number of IgA secreting cells was also increased under the same conditions detected by the ELISPOT assay. This specific activity of Sn was completely abrogated with the treatment of anti-TGF-01 antibody. The acid-activated Sn from the PP T cell fine contained 2.5 ng/ml of active TGF-01, assayed by using a MV 1 LU cell line. In addition, it was found that freshly isolated PP T cells synthesize TGF-(31 mRNA constitutively measured by using RT-PCR, indicating that TGF-(31 expression may be post-transcriptionally regulated by T cells. Finally, Sn from keyhole limpet hemocyanin-specific PP T cell line showed a similar effect on IgA production. The results from the present study suggest that TGF-01 can be expressed by intestinal PP¢¥T cells and the synthesized TGF-01 can induce IgA isotype switching in vivo.
