An Eight Base Pair Deletion Polymorphism in the Human L-myc Gene

Abstract
The human L-myc gene shows an EcoRI RFLP defined by a 10 kb or a 6 kb Ec+oRI fragment. A new polymorphic site, defined by the presence (N allele) or absence of an eight base pair (D allele) in the second intron of the L-myc gene was identified by comparing the nucleotide sequences of two chromosomes from a lung cancer patient (Lee et al., 1993). To investigate whether this deletion polymorphism is universal, nucleotide sequences of this region were deter-mined using DNAs from 14 individuals (5 N-N types, 5 D-D types, and 4 N-D types). Sequences were determined after amplification of this region by PCR and cloning of the amplified fragments. The sequences of N and D alleles from all samples were identical between the same alleles, suggesting that this eight base pair deletion polymorphism is universal.
