Posttranscriptional control of Light-induced apoprotein of Kight-harvesting Chlorophyll-protein complexes of Photosystem ¥± in Euglena gracilis Z

Abstract
Dark-grown Euglena gracilis Z were exposed to white fight which induces chloroplast development including a massive formation of thylakoid membranes. The temporal accumulation in greening cells of apoprotein of light-harvesting chlorophyll-protein complex of photosystem H (LHCP II) was quantitated by an immuno dot-binding assay. The temporal accumulation in the greening cells of the mRNAs for LHCP 1I was assessed by dot blot hybridization using 11P-labeled DNA probes. Dark-grown cells had a low level of apoprotein of LHCP U and high level of mRNA measured. Nuclear-coded 28 kDa LHCP U thylakoid protein accumulated after 6-12 h, the time chlorophyll began to accumulate. The comparison of the relative increase between mRNA and protein suggests that expression of the 28 kDa LHCP H gene(s) appears . subject to nuclear control, such as transcriptional control and/or stabilization of the transcript up to one day. After this, posttranscriptional control appears to be important. Overall, the fight-induced accumulation kinetics of the LHCP 11 and its mRNA shows that multiple controls, either overlapping or invoked at specific times, may regulate the synthesis of the protein during fight-induced chloroplast development in Euglena.
