Crystallization and Preliminary X-ray analysis of Probable Amylase/Protease Inhibitor-A from Rice Seeds

Abstract
Probable amylase/protease inhibitor-A (PAPI-A) from rice seeds, which shows a phospholipid transfer activity, was crystallized at 25 C in two crystal forms. Tetragonal crystals (form I) grew to dimensions of 0.5 mm X 0.4 mm X 0.4 mm using ammonium sulfate as a precipitant and diffracted to beyond 1.7 A Bragg spacing with X-rays from a rotating anode source. X-ray data have been collected to 2.16 A from a native crystal. The crystal belongas to the space group of P41212 or P43212 with unit cell parameters of a = b = 38.03 A and c = 98.11 A. Orthorhombic crystals (form 11) grew to dimensions of 0.5 mm X 0.4 mm X 0.1 mm using polyethylene glycol 400 as a precipitant and diffracted to beyond 1.8 A Bragg spacing with the same X-rays. Partial X-ray data have been collected to 2.6 A from a native crystal. The crystal belongs to the seaee group of either P222 or P2221 with unit cell parameters of a = 78.2 A, b = 54.5 A, and c = 49.9 A.
