The avian genome was thought to lack any retroelements except for several families of endogenous retroviruses. This work reports the isolation and the characterization of novel genetic elements which may be distantly related to the previously identified retroviruses and/or represent retrovirus-related sequences that are conserved in the line-0 chicken and other avian genomes. These elements were detected by polymerise chain reaction using pruners derived from a highly conserved region of the retrovirus reverse fimiscriptase gene and identified by sequence analysis. These novel families of endogenous retroelements are highly conserved in the avian genome, suggesting that they may be more ancient than any known avian endogenous retroviruses. They may represent primordial retroelements which might have been involved in the evolution of retroviruses.
