FK506 Inhibits Yeast FK506-Binding Protein-12 Fused to Maltose-Binding Protein

Abstract

|
The immunosuppressant FK506 prevents T-cell activation and also inhibits the growth of certain groups of yeast and fungi. It has been shown that yeast contains a 12-kDa cytosolic FK506-binding protein (yFKBP-12), which possesses a peptidylprolyl cis-trans isomerase activity. Using the polymerase chain reaction, 345 hp DNA encoding a FK506 binding protein-12 from yeast has been cloned, sequenced and expressed as a fusion protein with a maltose-binding protein. The fusion protein was purified by an affinity chromatography. By measuring the rate of chymotrypsin cleavage of the substrate N-succinyl-Ala-Ala-Pro-Phe p-nitroanifide, the fusion protein was found to have peptidylprolyl cis-trans isomerase activity comparable to that of a yeast FK506 binding protein. This peptidylprolyl cis-trans isomerase activity was inhibited by FK506.
