Expression and Cellular Localization of gonadotropin Releasing Hormone in the Rat Gonad

Abstract
The expression and cellular localization of GnRH transcripts and immunoreactive GnRH in the rat gonad were examined by in situ hybridization and immunohistochemistry, respectively. In the rat ovary, positive signals of GnRH transcript were predominantly shown in follicles and corpus luteum, while immunoreactive GnRH molecules were widely distributed in different cell types including interstitial, cumulus, granulosa, theca cells and corpus luteum. In the rat testis, GnRH transcript signals were found in discrete seminiferous tubules. Dense GnRH transcript signals were detected in the basal membrane, but not in lumen of seminiferous tubules containing mature sperms and Leydig cells. Immunoreactive GnRH molecules were widely distributed in Leydig cells, some spermatogenic cells and in most of the cytoplasm of Sertoli cells. This study clearly indicates that GnRH is locally synthesized in the discrete cell population of the rat gonad, supporting the notion that this peptide may exert autocrine and/or paracrine actions in intragonadal communication.
