Cloning and Functional Expression of the ddh Gene Involved in the Novel Pathway of Lysine Biosynthesis frpm Brevibacterium lactofermentum

Abstract
The Brevibacterium lactofermentum ddh gene encoding meso-diaminopimelate (meso-DAP)-dehydrogenase (DDH) was cloned by complementation of the Escherichia soli dapD mutation. Sub-cloning experiments and complementation tests indicated that the region for DDH activity was within the ^-2.5 kb XhoI fragment. Southern hybridization analysis confirmed that the cloned DNA fragment originated from B. lactofermentum genomic DNA. The activity staining showed that a specific band of DDH activity was observed in the strains carrying each subclone.
