Isolation and Characterization of STGA1, a Member of the TGA1 Family of bZIP Transcription Factors Soybean

Abstract
TGAI is a well-characterized transcription factor that may mediate the root-specific and auxin-responsive expression of some plant genes. The TGAIa was isolated from tobacco as an activation sequence factor 1 having nuclear DNA-binding activity observed in tobacco and pea. Since this factor interacts with sequences in the 75 region of the cauliflower mosaic virus 35 S promoter, designated as activation sequence 1, and many other cis-acting elements containing TGACG or related motifs, such as hex-1, nos-1, and other auxin-responsive elements, we studied soybean TGAI to address transcription regulation of gene expression from soybean seedlings. Here we describe the isolation and characterization of a soybean cDNA that encodes a soybean TGAI, named STGAI. Analysis of deduced amino acid sequences of the STGA1 cDNA revealed that the STGAI is a member of the basic leucine zipper (bZIP) class of transcription factor. The STGAI contains an N-terminal bZIP region as a DNA binding domain and C-terminal glutamine-rich region as a putative transcription activation domain. Northern analysis was performed to study expression of the STGAI gene showed that STGAI mRNA was most abundant in the mature section of hypocotyl and root tissues. The DNA-binding properties of cloned STGAI were also studied. The STGAI binds to many TGACG containing motifs including CRE (TGACGTCA), hex-1 (TGACGTGGC), and Aux28132 (TGACGACA), a cis-element present in the promoter of a soybean auxin-responsive gene GmAux28. This suggests that the STGAI is a good candidate for soybean ASF-1, interacting with TGACG-motif sequences of many soybean genes.
