Effects of a Mutation in the Discriminator Region of the rnpB P-1 Promoter on the Escherichia coli rnpB Gene Expression

Abstract
To examine the relevance of the discriminator region to the rnpB gene expression under different growth conditions, we constructed two E. coli lysogenic strains where the X prophages carried the IacZ gene fused to the wild type rnpB promoter and a mutant promoter, respectively. The mutant promoter had the TATT sequence in place of the CGCC sequence in the discriminator region adjacent to the rnpB P-I transcription start-point. (3-galactosidase activities synthesized from the wild type and the mutant rnpB promoter of the rnpB-lacZ fusion were determined in the lysogens grown at different growth media and different growth phases. The synthesis of P-galactosidase from either the wild promoter or the mutant promoter decreased during the transition of the exponential phase of growth to the stationary phase both in the rich medium and in the minimal medium. While the wild type promoter activity of the cells grown in the minimal medium was much lower than that of the cells grown in the rich medium, the mutant promoter showed no significant variation on the 0-galactosidase activity between the cells grown in the rich medium and in the minimal medium. The results indicate that the CGCC discriminator motif is involved in the regulation of the rnpB gene expression with different growth rates, but not at different growth phases.
