Effects of C©÷-Ceramide on Cell Cycle Alteration and C-fos Gene Expression in U-937 Cells

Abstract
The effects of C2-ceramide on cell cycle alteration and c-fos gene expression during differentiation of U-937 cells were studied. The treatment of U-937 cells with a cell-permeable C2-ceramide caused a 501/o inhibition of DNA synthesis. The analysis of cell cycle distribution showed that cells in the G1 phase greatly increased from 57/6 to 73% during a 72 h treatment of C2-ceramide (6 pM). At the same concentration of C2-ceramide, the induction of c-fos gene expression was observed in 15 min, peaked at 30 min, and decreased therafter. Cycloheximide (10 g/ml), the inhibitor of protein synthesis, also led to a 2.2-fold increase of c fos gene expression as compared with the control. These results implicate that C2-ceramide is the lipid mediator molecule involved in the differentiation of U-937 cells leading to the inhibition of DNA synthesis, cell cycle arrest at the G1 phase, and the induction of e-fos gene expression.
