Characterization of Genomic Locus of Yeast ASP3 Gene

Abstract
We have previously found that the 3¢¥ flanking sequences of the yeast ASP3 gene were the same as the flanking sequences of the 5S rRNA gene. To determine whether the ASPS gene is located near the 5S rRNA gene in the chromosome, we carried out Southern blot analyses with various ASPS gene probes. Southern blot analyses show that the ASPS gene is located adjacent to the rRNA gene clusters, and that another copy of the gene might he located at another region on the chromosome of X2180-IA strain. We also used a chromosome blotting method to identify the location of the ASPS gene in the chromosome. As a result, the ASPS gene was mapped to chromosome XV in the X2180-1A, L3262, and 20B-12 strains. The XE 223-1D strain appears to have lost the entire ASPS gene. Furthermore, another copy of the ASP3 gene was found on chromosome XII in the X2180-IA strain. These results imply that the ASPS gene was originally located on chromosome XV, but the gene was probably duplicated and moved to the flanking region of the 5S rRNA locus of the chromosome XH in the X2180-IA strain by an unknown mechanism.
