Clonal Variation in Cytokinin Contents of Crown Gall Cell Lines of Cytokinin-dependent Cultivar in Phaseolus vulgaris L.

Abstract
Crown gall tumors of Phaseolus vulgaris cv. Palgong induced by Agrobacterium tunu/itciens C58 were cultured in vitro and analyzed tier their endogenous cytokinins. Three tumor-originated cell lines which had grown well in hormone-free medium were selected by nopaline tests and Northern blot analyses. In the tumor-originated cell lines, cytokinin contents varied among three cell lines analyzed. Thus, zeatin was 1.8-3.7 g kinetin equivalents/kg callus tissue and zeatin riboside was 4.9-7.7 gg kinetin equivalents/kg callus tissue. Isopentenyl adenine and isopentenyl adenosine were not detected in any significant level. In the hypocotyl-derived non-transformed cell line, any significant level of cytokinin could not be detected as 1 nmole per kg callus tissue. This result may elucidate our previous reports that callus culture of Phaseolus vulgaris cv. Palgong was absolutely cytokinindependent and it should be directly related to the growth-limited production of endogenous cytokinin in this genotype.
