Ha-ras oncogene Inhibits the Mitogenic Stimulation of 12-O-Tetra-decanoyl-13-acetate in REF52 Cells and Does Not Affect c-myc Induction

Rat cell-line T24REF52 is a non-transformed REF52 clone which expresses T24 Ha-ras oncogene at 10-30% of endogenous protooncogene Ha-ras p21 [Franza et al., (1986) Cell 44, 4094181. By using T24REF52 cells, it was found that the Ha-ras oncogene inhibits the mitogenic effect of 12-0-tetradecanoyl-13-acetate (TPA). It was not the cytotoxic effect of Ha-ras oncogene, because T24REF52 cells mitogenically responded to a medium containing 10% fetal calf serum. As c-myc is frequently associated with cell proliferation, the c-myc induction pattern by TPA was observed by RNase hybridization assay. In T24REF52 cells c-myc was induced as normally as in control REF52 cells. Thus Ha-ras oncogene did not affect c-myc expression. We also observed that Ha-ras oncogene did not affect the protein kinase C activity.
