Induction of Apoptosis by SV40 Tumor Viral antigens and Its Blockade by Serum Survival Factors in Human Keratinocytes

Abstract
As E1A and c-Mye have been shown to cause apoptosis in serum free media, we have fested whether SV40 tumor viral antigens show the same apoptosis phenomena in serum-free media and whether it can be blocked by adding serum suuival factors, such as, insulin, transferrin, selenium, linoleic acid, and bovine serum albumin, which are normally present in the serum except mitogens, i.e., epidermal growth factor and insulin-like growth factor etc. We have used Ad12-SV4O hybrid virus immortalized human keratinocyte cell line RHEK-1 and its neoplastically transfurmed cell line RHEK-1/Ras. Indeed, SV40 tumor viral antigens cause apoptosis in the serum free media and the apoptosis phenomena can be blocked by adding serum survival factors which suggests that the presence of SV4O tumor viral antigens abrogate the necessity of serum mitogens for growth when the medium is supplemented with serum survival factors.
