Applicability of Fluorescently-tagged short Tandem Repeats Markers Genotyping for Forensic Application

Abstract

|
The genetic analysis of short tandem repeat markers based on (dC-dA)n(dG-dT)n dinucleotide repeat sequences using fluorescently-tagged primers were evaluated to the suitability for DNA typing including its accuracy, precision, and sensitivity. The average accuracy and precision of band-size estimates obtained by automated real-time analysis of fluorescently amplified products showed 99.48% and 99.83%, respectively. The minimal amount of target DNA required to detect a fluorescent signal on the Genescanner for short tandem repeat markers was approximately 0.04 ng of template genomic DNA, and allelic profiles could be obtained for cases of a highly degraded state of target DNA. Therefore, these facts shows that short tandem repeat genotyping by the automated analysis of fluorescently amplified products will be advantageous for forensic application.
