Light-regulated and Circadian Rhythmic Expression of a Potato Chlorophyll a/b Binding Protein Gene in Transgenic Tobacco Plants

Abstract
The maior light-harvesting chlorophyll a/b binding (CAB) proteins of the photosynthetic apparatus are encoded by a multigene family of nuclear cab genes in earh plant species. We previously isolated a type I cab gene (cab-14) of photosystem ¥± from potato. In order to elucidate its mode of expression, a chimeric gene was constructed with the 0.92 kb 5¢¥-flanking sequence of the cab-14 gene and chloramphenicol acetyptransferase (CAT) gene, and then the cab14-CAT gene was introduced into the genome of tobacco by Agrobacterium-mediated transformation. Expression of the chimeric gene was analyed by CAT assays with extracts of calli, mature plants and seedlings grown under darkness or light The cab14-CAT gene was expressed in a light-inducible manner in transgenic tobacco. The level of mRNA for the cab14-CAT gene was further analyzed under light-dark and under continuous light conditions by polymerase chain reaction with cDNAs synthesized by reverse transcriptase. The results indicated that its expression exhibited diurnal and circadian rhythmicity, reaching its maximum at about 9 h after illumination.
