Unusual Features of HIV-1 Infection of an Early Myeloid Cell Line

Abstract

We have previously described the infection of the early myeloid cell line, HL-6O, with a recombinant human immunodeficiency virus type 1 proviral clone, and reported that virus production from infected cells can be increased when the cells are induced to differentiate along the monocytic pathway by treatment with phorbol myristate [Cannon P. M., Tenen, D. G., Feinberg, M. B., Shin H. S., and Kim, S. (1993) Blood 81, 437-445]. We here report further investigations into the mechanism of this stimulation, noting that this does not occur at the level of transcription, as levels of viral RNA were unchanged fallowing PMA induction. In addition, we also observed a loss of infectivity of virus produced from these infected cell lines after several weeks in culture, despite an apparently normal morphology of the particles. This did not occur with a similarly treated T cell line, suggesting that it is a feature of human immunodeficiency virus infection of this early myeloid cell line.
