Analysis of the Coat Protein Gene of Odontoglossum Ringspot virus-Cy (Korean Isolate)

Abstract

|
Odontoglossum ringspot virus-Cy (ORSV-Cy), a Korean isolate, originally isolated from diseased leaves of a Korean native Cymbidium goeringii Reichenbach, was used for a source of virus. The purified virus was verified by electron microscopic obselvation of a virion particle and by SDS-PACE analysis of viral coat protein. Viral RNA was extracted from the purified ORSV particles and the viral RNA was polyadenylated in vitro at its 3¢¥-end. Double stranded cDNA was synthesized by using oligo(dT) primer and cloned in pSPORTl. The nucleotide sequence of ORSV-Cy comprises an open reading frame of 477 nucleotides, which encodes the virion coat protein of 157 amino acids with M_(r) 17,185. The sequences between the ORSV-Cy and two ORSV strains from Japan and Germany share the high degree homologies of 98.1% and 98.3% in the nucleotide and 96.2% and 95.6% in the amino acid lewels, respectively. At the amino acid level, the coat protein sequence of ORSV-Cy strain shares a range of 73.4% to 49.4% similar to those of other six tobamoviruses. ORSV is more closely related to tobacco mild green mosaic virus and pepper mild mottle virus than other viruses in the tobamovirus group.
