The Biochemical characterization of rice opaue Mutants

Abstract
Three rice endosperm mutants with opaque phenotypes, op-13, -14, and -17, were isolated. The opaque mutants are observed to be controlled by a single recessive gene according to the segregation analysis of phenotype. The characteristics of polymeric structures of starch in the mutant endosperm were analyzed using high performance liquid chromatography. Fractional analysis of starch after debranching with isoamylase showd that the total branching ratio was reduced due to the low ¡°A¡± branching ratio in the mutants. This result suggests that one of the two independent branching mechanisms is affected in the biosynthesis of amylopectin in the opaque mutants. In the mutants, ¡°A¡± branching system appears to be regulated by opaque mutations. Storage proteins in opaque and wild type seeds were analyzed using SDS-PAGE and two dimensional electrophoresis, revealing changes in the compositions of albumin and prolamin but not glutelin. Two 10 kDa prolamine and two 35 kDa albumin polypeptides are primarily expressed in opaque mutants, while two albumin polypeptides of 21 and 25 kDa are absent in the mutants. Taken together, our results suggest that opaque mutation in rice has a macrophenotype due to pleiotrophic effects on the biosynthesis of starch and proteins during endosperm development.
