Genetic Analysis of Interactions between arg-14 and arg-6 Gene products in Neurospora crassa

Abstract

|
Acetylglutamate synthase and acetylglutamate kinase, the first and the second biosynthetic enzymes for arginine in Neurospora crassa, are encoded by arg-14 and arg-6 respectively. Both enzymes are feed back-inhibited by the end product of the pathway, arginine. The second and the third enzymes of this pathway, acetylglutamate kinase and acetylglutamyl phosphate reductase are encoded by the single locus, arg-6 in N. crassa. Some arg-6 mutants lack acetylglutamate synthase activity or feed-back resistance. With these evidences, we proposed two models of the interaction between acetylglutamate synthase and acetylglutamate kinase and tested these models using genetic approaches in this study. The arg-6 mutant strains were mutagenized to obtain arg-6 pseudorevertants. Then, the linkage between intergenic arg-6 suppressor mutations and arg-14 was genetically mapped. Our data supported a model that a third protein is requird for protein-protein interaction beween the Arg-6 protein and the Arg-14 protein.
