Transformation of Methylotrophic Bacteria by Electroporation of pRO1727 Plasmid from Pseudomonas aeruginosa

Abstract
The transformation by means of electroporation of several methylotrophic bacteria with a covalently closed circular plasmid pRO1727 isolated from a non-methylotrophic bacterium Pseudomonas aeruginosa is described. 0.2-cm cuvettes were used with cells suspended in 10% (v/v) glycerol and Gene Pulser was set at 2.0-2.5 kV, 25 ¥ìF, and 200 ¥Ø giving a time constant of 4.5-4.8 s. Frequencies of transformation with cells growing at the late exponential growth phase varied from 10^(-3) to 10^(-6) per ¥ìg DNA, depending on the strain. The pRO1727 prepared from a methanol-utilizing bacterium was capable of being introduced back into the P. aeruginosa.
