Production of Infectious Viruses from Full-Length cDNA of Tobacco Mosaic Virus Korean Isolate

Abstract

A full-length cDNA copy of genomic RNA from a virulent tobacco mosaic virus strain isolated from Korea was constructed in an expression vector using bacteriophage T7 RNA polymerase promoter. The plasmid DNA was linearized at a unique restriction site down stream of the double-stranded cDNA insert. It was used as a template for in vitro transcription by T7 RNA polymerase in the presence of cap structure (m^(7)GpppG). The in vitro transcript synthesized was tested for infectivity on both local lesion and systemic hosts of tobacco plant. The typical necrotic lesions appeared on leaves of local lesion host inoculated with transcripts after 3 days. Inoculation of transcripts onto leaves of systemic host resulted in the accumulation of virus particles.
