Characterization of the Basal and Brain-specific Expression of the Human Hypoxanthine guanine Phosphoribisyltransferase Gene

Abstract

|
Mechanisms responsible for the expression of the human hypoxanthine guanine phosphoribosyltransferase (HPRT) gene have been studied. The 5¢¥ flanking region of the gene lacks the prototypical 5¢¥transcriptional regulatory elements such as TATA and CAAT boxes, but contains extremely GC-rich sequences and four putative Spl binding sites. DNA sequence elements required for the basal expression of the human HPRT gene were identified by measuring transient expression of the recombinant HPRT promoter-chloramphenicol acetyltransferase (CAT) gene transfected into mouse fibroblast cells. This analysis revealed that the 5¢¥-flanking region, located between - 220 and - 145 upstream from the translation initiation codon and containing the putative Spl binding sites, is essential for the basal expression of the human HPRT gene. Northern blot analysis showed that the high HPRT activity in brain tissue is correlated well with the increased steady state mRNA level in this tissue, suggesting that transcriptional regulation may play a role in the human HPRT gene expression in the brain tissue.
