Effect of Water Stress on total RNA , Polysome and Protein Changes during seed Germination of Cucumber (Cucumis sativus L .)

Abstract

Cucumber seeds were germinated in water potential of -0.24, -0.48 and -0.97 §ç with sorbitol or mannitol under light at 25¡É. The higher water stress resulted in the lower germination rate. In normal condition, the content of protein and total RNA were gradually increased as germination proceeded, but in high stress candition (-0.97 §ç), those were highest at the first day of imbibition and rapidly decreased afterwards. The content of polysome in -0.97 §ç plot was lower than that in control. The protein patterns were compared by SDS-PAGE. The patterns in seedling at control and -0.24 §ç plot showed similar band pattern. While the patterns of -0.48 and -0.97 §ç plot which showed restricted seedling growth showed similar protein patterns to that of dry seeds.
