Analysis of the Expression Pattern of Recombinant Proteins in vWF Transgenic Pigs

Abstract
We generated transgenic pig that produced recombinant human von Willebrand factor(rhvWF) in its milk using microinjection with bovine alpha S1-casein gene promoter controlled hvWF cDNA expression vector. Transgene was stably inserted into host genome from F0 to F3 generation of transgenic pig. This study is to analysis of expression patterns of hvWF in three lines. According to Western blotting and ELISA, the rhvWF expression of V3 line showed the highest level among three lines. Real time PCR and Southern blotting data showed that V1 had the largest copy number. To identify chromosome integration site, FISH and karyotyping were performed. Consequently, the transgene was located in chromosome 15q13.1 of V1, in chromosome 11p11.2 of V2, and in chromosome 4p13 of V3. Therefore, these results indicate that the expression level of vWF in transgenic pig was involved in the integration locus of chromosome.
