The extract of Tamarindus indica L. suppresses IKK¥â activity and NF-¥êB-dependent lymphoblastoid cell line survival

Abstract
The NF-¥êB family of transcription factors is a critical regulator for the cell survival by inducing the expression of anti-apoptotic genes. In this study, we found that a seventy percent ethanol extract of Tamarindus indica L. (TIE) strongly inhibits both IL-1¥â- and lipopolysaccharide (LPS)-induced IKK¥â and NF-¥êB activation without affecting the activity of IL-1 receptor associated kinase 1 (IRAK1), which functions upstream of IKK¥â. In addition, TIE had no effect on the LPS-induced c-Jun N-terminal kinase (JNK) or p38 mitogen-activated protein kinase (MAPK) activation. Furthermore, TIE clearly reduced the viability of NF-¥êB-dependent lymphoblastoid cell line (LCL) without adversely affecting the viability of NF-¥êB-independent HeLa cells. Thus, TIE targets IKK¥â to inhibit NF-¥êB activation and LCL viability.
