Effect of Cudrania tricuspidata Water Extract on TNF-¥á-Induced Endoplasmic Reticulum Stress and Changes in Insulin Signaling and Lipid Metabolism in HepG2 Cells

|
Hepatic inflammation and endoplasmic reticulum (ER) stress are associated with several metabolic conditions, including obesity and hyperglycemia. We evaluated the effects of the water extract of Cudrania tricuspidata leaves (CTL) in the tumor necrosis factor-alpha (TNF-¥á)-treated HepG2 cells to examine its protective effects in TNF-¥á triggered ER stress, insulin resistance, and lipogenesis in hepatic cells. The TNF-¥á-stimulated control group showed a marked increase in the inflammatory cytokines [TNF-¥á, interleukin (IL)-1¥â, and IL-6] and ER stress-related factors (IRE1¥á, eIF2¥á, ATF4, CHOP, XBP-1, and GRP78). Additionally, TNF-¥á suppressed the insulin signaling IRS/Akt pathway and increased the mRNA expression of SREBP-1 and FAS. The CTL treatment, however, caused the interruption of TNF-¥á-induced inflammation and ER stress in the HepG2 cells. Moreover, the treatment with CTL resulted in the activation of insulin signaling and inhibition of gluconeogenic gene expression and lipogenesis. In conclusion, these results suggest that CTL can protect against inflammation as well as ER stress, insulin resistance, and lipogenesis in the liver and that it may be a useful agent in the treatment of hyperglycemia.
