Establishment of Stem Cell Containing System Using Mouse Intestine

Abstract
Stem cells (SCs) have ability to perpetuate themselves through self-renewal without changing their physiological, molecular biological characters and to generate fully differentiated functional cells. Since their properties, SCs may have the potential to be used as a developmental model, a source for pharmaceutical screening and cell therapy for tissue regeneration. To investigate the in vivo characterization of them, we designed the in vivo cell containing system using mouse. About 1.5 §¯ long segment was separated from small intestine of rodents. The segment should have intact blood supply with mesenteric artery. The separated segment was inverted inside out, and the mucosal layer was denuded using dental curette. Then, the segment was restored and one million GFP expressed liver cells were transplanted. Finally, the segment containing transplanted cells was anchored to peritoneum. Animals were sacrificed at variable times after surgery. To demonstrate existence of transplanted cell, we carried out histological examination. Histological examination showed complete removal of intestinal mucosa and the transplanted cells appeared healthy. Operated animals lived for 10-30 days. Most intestinal segments were intact and the transplanted cells appeared alive in the segment. These results indicate that bio-containing system using intestinal segment can be a way of long-term culture system in vivo. Therefore, the establishment of bio-containing system may be helpful for basic stem cell research and tissue regeneration.
