Neuroinflammatory Activation is Reduced by Intracerebral Transplantation of Embryonic Cerebellar Cells in Niemann-Pick Type C Mice

Abstract

|
Recent studies have shown that bone marrow derived-mesenchymal stem cells (BM-MSCs) improve neurological deficits when transplanted into animal models of neurological disorders. Cell replacement after neuronal degeneration in the adult central nervous system (CNS) depends on availability of specific cues to direct differentiation, specification and integration of newly born neurons into mature circuits. Following recent reports indicating that neurogenic signals may be reactivated in the adult injured CNS, here we asked whether embryonic cerebellar cells transplanted to the cerebella of adult neurodegenerative Niemann-Pick disease type C (NP-C) model mice. Most notably, there was a higher amount of Purkinje neurons and reduced significantly both of astrocytic and microglial activations in the cerebellum of embryonic cerebellar cells transplanted NP-C mice. These findings suggest that embryonic cerebellar cells transplantation may have potential for a therapeutic role in the treatment of NP-C and other neurodegenerative brain disorders.
