Development of DNA Chip for the Differential Diagnosis of Marek¡¯s Disease and Avian Leukosis/Sarcoma in Chickens

Abstract
Chicken lymphomas caused by either Marek¡¯s disease virus (MDV) or avian leukosis/sarcoma virus (ALSV) were fastidious diseases for the differential diagnosis using ordinary methods including histopathology and PCR assays. Therefore, the aim of this study is to develop DNA chip for the differential diagnosis of Marek¡¯s disease (MD) and avian leukosis/sarcoma (ALS) in chickens. Using the primer pairs specific to each serotype of MDV and each subgroup of ALSV, single PCR assays were carried out with DNA extracted from each prototype strain of MDVs and ALSVs and amplified each specific band for prototype strains of MDVs and ALSVs. DNA chip assays were performed with the products by multiplex PCR assays using mixture of primer pairs specific to each MDV serotype and each ALSV subgroup virus. The positive reaction of each serotype of MDV and each subgroup of ALSV was detected on the DNA chip slide. DNA chip assays also detected MDVs even in negative by RT-PCR assays. From these results, the DNA chip assays developed promised accurate and rapid diagnosis for the differentiation of MD and ALS in chickens. This is the first report to detect MDVs and ALSVs by DNA chip assays in the world.
