Hermaphroditism occurs rarely in human and animal populations. Previously, we reported one hermaphrodite case and also found other three reported cases in Korea. Furthermore, recently, we found other two cases of hermaphrodite dog. In this study, we analyzed the characteristics of the submitted cases and aimed to differentiate their sexing using a molecular analytical method. All hermaphrodites were identified in American Cocker Spaniel dogs in Korea. The characteristics were its enlarged os clitoris that was shown a reddish finger-like structure protruding from the vulva. The dogs revealed vulva hypertrophy, perivulvar dermatitis, skin fold dermatitis and clitoris hypertrophy. The dogs were diagnosed as hermaphroditism with ovotestes. As the results of molecular analytical sexing, two submitted dogs were identified as female. To our knowledge, hermaphroditism may be a rare case in dog. However, as the results of this study, American Cocker Spaniel dogs have susceptible to hermaphroditism. We concluded that these findings contributed to research a genetic sexual difference disturbance syndrome like as hermaphroditism. Also, PCR revealed a very efficient tool for animal sexing.
