A Cell Culture System for Mouse Embryonic Neuronal Progenitor Cells: Expression of Metabotropic Glutamate Receptor, mGluR2/3

|
The isolation and in vitro differentiation of neural progenitor cells (NPCs) provide an approach to study the regulation of development and differentiation in cell culture. This paper describes a culture system of embryonic-derived NPCs. NPCs from mouse cortex of embryonic day 14 (E14) can be maintained and expanded using a serum free defined medium containing basic fibroblast growth factor (bFGF). In the highly purified secondary cultures, most of these cells were positive for nestin, a specific marker for NPCs in developmental brain. Immunocytochemistry analysis showed the expression of metabotropic glutamate receptor, mGluR3 in the nestin-immunoreactive cultured embryonic-derived NPCs. Over 99.7% of the nestin-expressing cells co-expressed mGluR2/3. These results suggest that mGluR3 may play a role in the early neurogenetic events. Here, this method will facilitate studies using embryonic-derived NPCs as cell culture system for neural development and differentiation.
