The Antinociception of BD1047 and its Synergy with Other Analgesics in Visceral Pain Model

Abstract

There is evidence supporting the antinociceptive effects of sigma-1 receptor antagonist, BD1047 in various models of neuropathic pain as well as inflammatory somatic pain. However, data are lacking on the antinociceptive potential of BD1047 against visceral pain. The aim of present study was to determine the antinociceptive effect of BD1047 in the model of visceral acute tonic pain, the acetic-acid writhing test in mice. Furthermore, present study was undertaken to characterize the interactions between BD1047 and other analgesic candidates. Intrathecal pretreatment of BD1047 (10, 30 and 100 n§ß/mice) produced dose-dependent antinociceptive effect against acetic acid-induced writhing. Intraperitoneal injection of several analgesics including fluoxetine, paroxetine, nisoxetine, morphine, ketamine and acetaminophen dose-dependently produced anti-nociceptive effect in acetic acid induced pain model. Combination of low effective doses of these analgesic drugs with BD1047 (30 n§ß/mice) resulted in synergistic interactions. This result may be indicated that spinal blockage of sigma-1 receptor activity evokes synergistic interaction of these analgesic drugs in visceral pain model.
