An experimental study on the effects of Sagan-Tang, Bogan-Tang upon the Liver damage in Rats



Abstract
On the assumption that the hepatic dysfunction itself can be regarded as a condition of fullness symptom when Sagan-Tang was found to be effective upon the administration of 3ogan-Tang and Sagan-Tang to the animal with hepatic damage due to Ally! alchol, changes in the activities of GOT, GPT, ALP, LDH and ACP, often applied for the determination of hepatic function, were determined to observe biochemical as well as histochemical changes and the following results were obtained.
1. Sagan-Tang brought about significant changes (P < 0.05) in the activity of GOT at 36 hours (196.0 22.9) and at 48 hours (152.5 29.1) after the administration of liver damaging substance in contrast to the case of Bogan-Tang which caused a significant change (P < 0.05) only once at 30 minutes (92.0 7.3) after the administration. Significant changes (P < 0.05 and P < 0.02) in the activity of GPT were observed at 48 hours (67.0 10.3) and 60 hours (65.5 9.5) after the medication for the case of Sagan-Tang whereas a significant change (P < 0.05) in the activity of this enzyme at 30 minutes (43.0 12.1) was noted in the case of Bogan-Tang.
2. As for the change in the activity of ALP of drug administered animal, Sagan-Tang demonstrated significant differences (P < 0.05) at 60 hours (15.0 2.1) and 72 hours (11.0 1.0) whilst Bogan-Tang, ineffective.
As for the enzymatic activity, a mediocre positive reaction (++) similar to that of normal was observed between 36 to 72 hours of medication was observed in the case of Sagan-Tang whereas a continuous slightly positive reaction (+) was recorded from 30 minutes to 72 hours in the case of Bogan-Tang.
3. Sagan-Tang caused a significant change (P < 0.05) in the activity of LDH of
animal administered with antihepatic drug at 60 hours (400.0 42.6) but Bogan-Tang did not cause such a change. A mediocre positive reaction(++) of enzymatic activity somewhat different to that of normal was noted in the case of Sagan-Tang during 24 to 72 hours but a lesser positive reaction(+) was continuously observed during 30 minutes to 72 hours in the case of Bogan-Tang.
4. The activity of ACP following the drugging was significantly (P < 0.05) changed at 60 hours (6.0 1.0) and 72 hours (4.0 0.5) by Sagan-Tang but it was not significantly changed by Bogan-Tang. A mediocre positive reaction (++) of the enzymatic activity similar to that normal was observed between 36 to 72 hours under the influence of Sagan-Tang in contrast to the case of Bogan-Tang in which a continuous slight positive reaction(+) was registered during 30 minutes to 72 hours,

In view of the above conclusions, Sagan-Tang can be said to have a recuperative action on the increased activity of enzymes following the histological changes of liver ranging from blood congestion inflamation to necrosis.
Furthermore, in view of the traditional prescription of Sagan-Tang in oriental medicine for a series of syndrom expressed as blood fullness or excessiveness of blood, the histological change due to hepatic dysfunction can be regarded as a model of hepatic fullness syndrome.
