Histological Studies on the effect of the Deer Horn Water Extract on the Thyroid Gland of Straved Rat


Abstract
It is well known that the deer horn (Cervi Cornu) is a restorative and hematopoietic agent in Oriental Medicine. This study was carried out to clarify the effect of deer horn water extract on the recovery of morphological ogical changes of thyroid gland as a result of hypothyroidism induced by acute starvation.
Adult male rats (Sprague - Dawley strain) weighing about 200g body weight were subjected to acute starvation for periods of 7 days by complete withdrawal of the basic normal ration. After acute starvation, the experimental animals were divided into two groups and refed. The duration of the refeeding periods were one, two, four, eight and sixteen days.
The one (BF) group were refed with basic normal ration alone, the other (DF) group refed with basic normal ration plus a daily supplement of 0.2m1 deer horn water extracts perg body weight. At the conclusion of the each refeeding period the animals were killed, the thyroids removed and fixed in 10% neutral formalin solution. The one were imbedded in paraffin, sectioned and stained with hematoxylin - eosin and periodic acid Schiff¢¥ (PAS) -- Hematoxylin.
The following results were obtained:
1) In animals starvation for 7 days, the diameter of follicle in the thyroid gland were 40 - 100um. Follicular epithelium repersented simple squamous epithelium and colloid showed weak positive with PAS - reaction.
2) In animals refeeding for 1 and 2 days, the morphological change of thyroid gland
in,BF and DF - group showed to be the same as starved rats.
3) In animals refeeding for 4 and 8 days, the histological fmdings of BF - group were similar to that seen in the starved rats. But the diameter of thyrioid follicles in DF - group were 20 - 80um and follicle were lined with simple cuboidal epithelium. Colloid were appeared strong positive with PAS - reaction as normal control animals.
4) In refeeding for 16 days, the structure of thyroid gland in BF and DF - group were repersented normal appearance.
