Effects of Cervi cornu on Depression of Immune Response Induced by Methotrexate in Mice


Abstract
In order to verify the anti-pathogenic factor (1) and pathogenic factor (iJ13) in Oriental Medicine, the author investigates the effects of Cervi cornu (CC) on depression immune response induced by methotrexate(MTX). ICR mice having a body weight of about 20g are used as experimental animals dividing them into four groups-Saline, CC, MTX, CC+MTX group.
All of the mice were sensitized i.v. with 108 sheep red blood cells(SRBC) and challenged i.d. with 108 SRBC 4 days later. Such immune responses as delayed-type hypersitivity(DTH), rosette forming cells (RFC), hemagglutinin titer (HA titer) and hemolysin titer (HL titer) were measured at 24 hours after challenge.
The results were as follow:
1. DTH was significantly increased in CC group. as compared with Saline group and in CC+MTX group as compared with MTX group.
2. RFC was significantly increased in CC group as compared with Saline group and in CC+MTX group as compared with MTX group.
3. HA titer was significantly increased in CC group as compared with Saline group and in CC+MTX group as compared with MTX group.
4. HL titer was significantly increased in CC group as compared with Saline group and in CC+MTX group as compared with MTX group.
From the above experimental results, Cervi cornu enhance the cell-mediated and humoral immune response, and it restrains the depression of cell-mediated and humoral immune response induced by MTX. Hence it is the conclusion of the author that the insufficiency of the anti-pathogenic factor 0E10 in Oriental Medicine is very similar with Depression of Immune Response in Occidental Medicine.
