Effects of Ginseng Radix Aqua-acupuncture on Immune Suppression Induced by Methotrexate in Mice



Abstract
In order to investigate the effects of Ginseng Radix aqua-acupuncture on immune suppression induced by methotrexate (MTX) , MTX was admini stered and aqua-acupuncture of Ginseng Radix into the loci corresponding Choksamni (S36) and Chung¢¥wan (CV12) of the human body to mouse was done for 3 days, and then the mice were sensitized I,v,with 108 sheep red blood cells (SRBC), and thereafter challenged i , d _with 108 SRBC on the 5 th day. Cell-mediated immune response for delayed type hypersensitivity (DTH) , rosette forming cells (RFC) and humoral immune response for hemagglutinin (HA) titers, hemolysin (HL) titers were measured at 24 hours after challenge. Phagocytic activity of reticuloendothelial system(RES) was studied by using the carbon clearance in ICR mice
The results were summarized as follows :
1. DTH was increased with statistical significance in aqua-acupuncture of Ginsing Radix on Chung¢¥wan and Choksamni loci but in case of Blank locus valuable increase of DTH was not recognized as compared with MTX administered group.
2. RFC were increased remarkably in aqua-acupuncture of Ginseng Radix on Chung¢¥wan locus and on Choksamni loci but in case of Blank locus any significance was not appeared as compared with MTX administered group.
3. HA titers were increased remarkably in aqua-acupuncture of Ginseng Radix on Chung¢¥wan and Choksamni loci but there was no valuable increase on Blank locus in comparison with MTX administered group.
4. HL titers were increased significantly in aqua-acupuncture of Ginseng Radix on Chung¢¥wan and Choksamni loci but there was no significant increase on Blank locus in comparison with MTX administered group.
5. The phagocytic activity of reticuloendothelial system was increased only in aqua-acupuncture on Chung¢¥wan locus as compared with MTX administered group.
According to the above results, the aqua-acupuncture of Ginseng Radix on Choksamni and Chung¢¥wan loci enhances the cell-mediated and humoral immune response suppressed by MTX in mice and also increases the phagocytic activity of reticuloendothelial system. Thus, it is suggested that aqua-acupuncture of Ginseng Radix could be recommended for the treatment and prevention of disease by virtue of improving host defence mechanism
