Study on Anticancer Effects of Putative Ponciri Fructus, Houttuyniae Herba, Manitis Squama and Polyporus in Rats


Abstract
Previous studies have demonstrated that the extracts of Ponciri Fructus(P.F.) and Houttuyniae Herba (H . H) augment natural killer cell activity (NKCA) in rat in vitro, and that tumor cells were sensitive to the extracts of H.H., Polyporus (P . P) and Manitis Squama (M.S.) . This study was performed to reveal the in vivo antitumor activity of the above 4 herb drugs.
At first, the extracts of P.F. and H.H. were administrated to CBA/NJ.0 mice orally or intraperitoneally (I .p) for 3 weeks, and then NKCA was tested. No differences were observed between the NKCA of control and treated groups. The mice were also inoculated i,p with Sarcoma-180 cells and 6 days after NKCA were tested and survivals were checked. NKCA of all groups declined as compared with those before tumor injection. However, NKCA of treated groups with P.F., i , p and H.H orally were significantly higher than that of control group (P<0
05). Percent survival at day 35 were 0.0%, 50.0%, and 66.7% in control, P.F.-treated, and H H.-treated group, respectively.
In second experiment, mice were inoculated with Sarcoma-180 cells and followed by treatment with extracts of H.H. P.O. and M.S. Mean survival time (MST) of control gruop was 18.8 days, and the injection of H .11. extract prolonged the MST to 27.8 days (P<0.05) . The MST of the mice treated with P.P. was prolonged to 27.8 days without statistical significance.
Taken together, these results show that the extracts of P.F. and H.H. contain antitumor substances, and suggested that such effects of P.F. and low dose of H .11. extract might result from NK cell activation. And it was considered that the antitumor activity of the 2 herb drugs fluctuated according to the dose and route of administration
