Experimental studies on Samulatang and herbal compositions in Samultang

Abstract
To investigate interactive effects of each herb in Samultang, Angelicae gigantis Radix, Cnidii Rhizoma, Rehmanniae Radix Preparat, and Paeoniae Radix Alba, test liquids, which have one of them, three ones of them, and all of them, were used to examine pH and portions of wet substances of the liquids; and difference in extractive rates of paeoniflorin, contractive antagonistic phenomenon of an isolated ileum, inhibitory effects of enzymatic activity in ¥â-glucuronidase, ¥á-g1ycosidase, and trypsin, and a inhibitory effect of heat induced hemolysis in vitro state: and a pain relieving effect by the acetic acid method, a sleeping time extension effect by pentobarbital-Na in vivo state, a bleeding time extension effect, an anti-thrombotic effect, and blood pressure and heart rate in SHR.
The results were summerized as follows ;
1. Among pH¡¯s of the extracts, Rehmanniae Radix Preparat revealed a strong acidity of 4.07 and all the combinations including Rehmanniae Radix Preparat and Samultang showed a week acidity.
2. The portions of wet substances were found to increase in the combinations without Rehmanniae Radix Preparat.
3. The extraction rate of Paeoniflorin in Paeoniae Radix Alba increased significantly when the combinations Angelicae gigantis Radix & Paeoniae Radix Alba.
4. When the isolated ileum was contracted by acetylcholine chloride, barium chloride, and histamine hydrochloride, it was found to be an antagonistic effect between the herbs.
5. Each of Paeoniae Radix Alba and Cnidii Rhizoma was found to inhibit considerably the activity of ¥â-glucuronidase, and antagonistic effects between the herbs were observed.
6. Each of Paeoniae Radix Alba and Rehmanniae Radix Preparat showed a remarkable inhibitory effects of the activity of ¥á-glycosidase, and antagonistic effects between the herbs were inspected.
7. Each herbs showed inhibitory effects of the activity of trypsin to their porpotion concentrations, while only the combinations including Cnidii Rhizoma showed synergic effect between the herbs and the other combinations and Samultang displayed an antagonistic effect between the herbs.
8. A inhibition effect appeared in the heat induced hemolysis by Angelicae gigantis Radix, Cnidii Rhizoma, and Rehmanniae Radix Preparat, and antagonistic effects between the herbs were inspected.
9. In the pain relieving experiment by the acetic acid method, each herb and the combinations showed pain relief effects, the combinations with Angelicae gigantis Radix & Paeoniae Radix Alba and Samultang were found to have considerable synergic effects.
10. No significant result was detected in the sleeping time extension experiment using pentobarbital-Na for each herb, but the combinations with Angelicae gigantis Radix & Paeoniae Radix Alba and Samultang were found to have considerable synergic effects.
11. Bleeding time extension effects appeared notably in Angelicae gigantis Radix, Paeoniae Radix Alba, and Cnidii Rhizoma, in addition the combinations without Rehmanniae Radix Preparat and Samultang exhibited considerable synergic effects.
12. The anti-thrombotic experiment indicated that Paeoniae Radix Alba have a remarkable coagulation time extension effect and the combinations with Paeoniae Radix Alba and Samultang make the coagulation time extend, suggesting that Paeoniae Radix Alba plays a main role for an anti-coagulation effect.
13. From the blood pressure and heart rate in SHR experiments, it was discovered that Angelicae gigantis Radix and Cnidii Rhizoma show considerable depression effects, and the combinations with Cnidii Rhizoma and Samultang also show the effects, proposing that Cnidii Rhizoma is the principal herb to give the effects : Angelicae gigantis Radix and Samultang can slow down the heart rate.
