Experimental Study On Tissue Response Of Different Demineralization Method


Abstract
Bone graft in the field of oral and maxillofacial surgery is being used in congenital deformities, trauma, infection & acquired deformities followed by tumor resection, and plastic augmentation of lowered alveolar ridge height.
Demineralized bone is manly composed of organic substance since it is prepared by removing inorganic component of apatite in bone and can be used in various configurations. The mechanism of bone formation after grafting demineralized bone is
characterized by cartilage formation followed by subsequent bone formation as in endochondral bone formation induced by osteogenic protein within matrix of demineralized bone.
Rejection occurs against untreated allogeneic bone of which the most cellular components are known to possess graft antigen. The major provoking factor of this immune reaction was confirmed to be subcellular particle similar to DNA in bone. In
order to
avoid stimulation of host immune reaction and reduce immunity of graft material, a variety of methods are recommended such as heating, irradiation, simple freezing and freeze-drying so that allogeneic bone graft doesn't stimulate immune response
of
host
tissue.
The purpose of this study was to compare simple demineralized bone power with freeze-dried demineralized bone powder in the capacity of healing and bone induction. Demineralized bone powders in the range of 500 - 1000§­ particle size were grafted
into
femur of rabbit and the healing processes were observed histologically through light and electron microscope.
@ES The results were as follows ;
@EN 1. Fibrous capsule with irregular resorption pattern around grafted bone was observed after 1 week in both group.
2. After 2 weeks, the degree of inflammatory cell infiltration in the experimental group was observed to be less than that of control group while the amount of new bone formation and osteoblastic apposition were relatively more active than
control
group.
3. Somewhat large cells of chondrocyte-like appearance were observed in both experimental and control group from the 2nd week of experiment.
4. While the overall histological feature of the 3-week experimental group showed similar patten to that of 4-week control group, activity of grafted bone matrix was great and the appearance of osteoclast was sparsely observed,.
5. In EM feature of the 4th week, processes from cell body of osteocytes in lacunae were found to be extended between canaliculi.
6. From the above mentioned results, it might be concluded that freeze-dried demineralized bone is better in the aspects of new bone formatio than simple dried demineralized bone.
