In Vitro Study On The Cytotoxicity Of The Differently Sterilized Demineralized Bone Powder


Abstract
All the process, acquiry, demineralization, cleansing etc, of demineralized allogeneic bone must be conducted under a germ free stable condition without bacteria and/or viruses contamination. But this sterilization process is very difficult. Even
though
the bone is acquired under a germ free state, we must have confidence of disinfection of all the solutions that come in contact with tissue and blood. To make germ free allogeneic bone materials, many kind of antibacterial agents have been
introduced,
for example, Merthiolate, formaldehyde, glutaraldehyde, alcohol, formalin, and acetone. Recently ethylene oxide gas sterilization or radiation sterilization is being frequently used. The hiological and biochemical response of the graft material
differs
with the type and concentration of the disinfectant, and various toxic reactions have been reported due to the graft material itself and the substance released by the disinfectant.
The author conducted the Millipore filter test, which is a toxic test on fibroblast according to the effect on activity of succinate dehydrogenase, to confirm the cytotoxicity of sterilization with irradiation and ethylene oxide gas to the
demineralized
allogeneic bone powder.
@ES The results were as follows:
@EN 1. Around the copper disk, positive control group, 10mm diameter discoloration could be seen.
2. As same as the negative control group, the silicone disk showed no discoloration.
3. The demineralized allogeneic bone which was sterilized with ethylene oxide gas or irradiation showed no cytotoxicity.
4. From this results, it is suggested that treatment with ethylene oxide gas and irradiation should be effective to sterilized the demineralized allogeneic bone.
@ES The results were as followings ;
@EN 1. Male : Female ratio was 17:23 and the 32 patients(80%) were twenties.
2. 26 patients(65%) complained not only esthetic problems but also functional problems, only esthetic problem(10, 25%), and only functional problem(4, 10%) were found.
3. Mandibular prognathism was found most frequently(25, 39%) and facial asymmetry(13.20%), angle hypertrophy(11. 17%) were found also.
4. Sagittal split ramus osteotomy was done most frequently(27, 35%), and LeFort I osteotomy(13, 17%), Angle reduction(12, 16%), Genioplasty (11, 15%) were done also.
5. Cases of nonspecific etiologic factor(22, 55%), cases of inherited tendency(12, 30%), cases of congenital anomaly(3, 7.5%), and cases of trauma(3, 7.5%) were found.
6. As analysis of operation site, cases of only maxillary surgery(2, 5%), cases of only mandibular surgery(23. 57.5%), and cases of simultaneous two jaw surgery (15, 37.5%) were found.
