Acrosome Reaction of Mouse Sperm by Human Follicular Fluid

Abstract

|
This study aimed to evaluate the acrosome reaction (AR) inducing activity of human follicular fluids (hFF) according to their estrogen (E2) and progesterone (P4) contents. When 1% (v/v) hFFs were treated to mouse sperm preincubated for 120 min,
AR
was
variable according to source of hFF. E2/P4 molar ratio of hFF rather than quantities of steroids (estradiol and progesterone) was positively correlated with AR. HFF depleted their organic solvent-soluble fraction showed reduced AR inducing
activity
when
it was treated to sperm by 10% (v/v). It suggested that organic solvent-extractable substances in the hFF might be responsible for AR inducing activity but water soluble phase of hFF is also important for AR induction. This result suggested that
E2/P4
molar ratio reflected physiological status of hFF responsible for AR inducing activity and can be used as a criterion to select hFF for sperm treatment in assited reproduction. In conclusion, mouse sperm AR test can be used as a simple, rapid,
and
discriminating method to select follicular fluid for sperm treatment in assisted reprodution.
