Studies on oral toxicity of Inchinchunggan-tang in mice

Abstract
This study was carried out to investigate the oral toxicity of the Inchinchunggan-tang in ICR mice.To investigate acute oral toxicity. 5,000, 2.500. 1,250, 625 and 312.5mg/kg of the Inchinchunggan-tang was given orally once to mice and symptoms and several items such as body weight, daily food and water consumption, gross findings of each organs were observed.To investigate subacute-chronic oral toxicity. 4.000 mg/kg(G1). 1.600mg/kg(G2) and 640mg/kg(G3) of the Inchinchunggan-tang was given orally to mice once a day for 28days and several items such as hematologic and serologic values were examined and following results were obtained.
1. Inchirchunggan-tang, administered both for once and 28 continuous days, did not induce any toxicological effect in the mortality in both sexes of mice.
2. Inchinchunggan-tang, administered both for once and 28 continuous days, did not induce any abnormal symptoms and signs.
3. Inchinchunggan-tang, administered for 28 continuous days, induced weight gain in male G2 and female GI groups with P(0.01. P(0.05, respectively.
4. Inchinchunggan-tang, administered both for once and 28 continuous days, did not induce any differences in food and water consumption between experimentaland control groups.
5. Inchinchunggan-tang, administered for 28 continuous days, did not induce any abnormal findings in opthalmoscopy, and the results of the unrinalysis were not significant.
6. Inchinchunggan-tang, administered for 28 continuous days, induced increase of the serological values such as AST. ALT and ALP leve!s in female mice G1 group(P(0.01), and ALP levels in female G2 and G3 groups(P(0.05, P(0.02, respectively).
7. Inchinchunggan-tang, administered for 28 continuous days, induced significant decrease or increase of the hematological values such as RBC, Hgb, hematocrit, MCV. MCHC, PCT, PLT levelS(P(0.01) in male G1 group and RBC. MCHC. MPV levels(P(0.01) and Hgb, MCV levels(P(0.05) in male G2 group.
8. Inchinchunggan-tang, administered both for once and 28 continuous days, did not induced any toxicological effect in the gross findings in boih sexes of mice.
9. Though both Inchinchunggan-tang group, administered for 28 continuous days, and control group showed histopathological abnormalities such as interstitial cell infiltration and tubular degeneration of the kidney, extrarnedullary hematopoiesis in spleen and liver, and congestion and thickening of alveolar wall in lung, it was riot consistent with the dose of the Inchinchunggan-tang.
According to the above result, it is considered that the Inchinchunggan-tang does not induce the acute or subacute-chronic toxicity in mice and is safe when administered with general dosage in clinical use.
