Effect of Oryungsan(¿À·É»ê) on the histoligical Changes in liver and epididymal fat and the biochemical changes of lipid metabolism and urine hornmone changes in obese rats induced by high fat diet




Abstract
The purpose of this study
This experimental study was designed to investigate the effects of Oryungsan on the weight and histological changes in liver, epididymal fat and lipid metabolism and urine hormone in obese rats induced by high fat diet.
Methods
Total population of sixty male rats(Sprague-Dawley) were utilized for this investigation. All rats selected for this study were ten weeks old with weight of 2505g. The first group of twenty rats(Nonnal group) were fed with normal diet(Samyang Co. Korea), second group of twenty rats(Control group) were fed with high fat diet(Harlan, TD94095, USA), and the remaining twenty rats(Sample group) fed with high fat diet and the Oryungsan. The measurement has been performed on (1) the weight of epididymal fat pad with electric weight gauge(Librorel-1200H, Shimadzu Japan) (2) the average size and the number of epididymal fat cells with light. microscope (Olympus, Japan) and Computer Image Analysis System(Image Analysis System, Leica Q500MC:, German) (3) the area % of fat drops in hepatic lobule with light microscope and Computer image analysis system (4) the quantity of total cholesterol in serum on enzyme-- method C-Allian system (5) the quantity of triglyceride in serum on enzyme method Van Handel system (6) the quantity of free fatty acid in serum on enzyme method Noma A system (7) the quantity of total lipid in serum on Sulfo-phospho-vanillin method (8) the quantity of phospholipid in serum on nzyme method (9) the quantity of HDL-cholesterol in serum on enzyme method (10) the quantity of catecholamine in urine on HPLC method external standard system. Zesults
1. It showed significant evidence that high fat diet conjunction with Oryungsan decreased the .veight of rats compare to control group.
2. It showed significant evidence that high fat diet conjunction with Oryungsan decreased the epididymal fat pad weight, the average size of epididymal fat cells and the number of epididymal fat cells compare to control group.
3. It showed significant evidence that high fat diet conjunction with Oryungsan decreased the area % of fat drops in hepatic lobule compare to control group.
4. It showed significant evidence that high fat diet conjunction with Oryungsan decreased the total cholesterol, triglyceride, free fatty acid and total lipid, phospholipid in serum compare to control group. It increased HDL-cholesterol in serum compare to control group.
5. It showed significant evidence that high fat diet conjunction with Oryungsan decreased epinephrine in urine compare to control group.
Conclusion
Based on these results, it is proved that Oryunsan decreased the weight of fat, the size of fat cells and the number of fat cells. So it is concluded that the clinical application of Oryungsan can help the treatment of obesity.
