The Effect of Dangkijakyaksan(Ó×ÏýíËå·ß¤) on Female Sex hormones and Cerebral atrophy following Middle Cerebral Artery Occlusion in Rats

Abstract
The purpose of this study is to find out the effections of Dangkijakyaksan( 15V_ft) on female sex hormones and cerebral atrophy after middle cerebral artery occlusion in female rats.
METHODS :
In this experiment, we used 26 Sprague-Dawley rats, weighing 190-200g. Cerebral infarction was induced from subtemporal method that is modified by Chen. Then we electroc:auterized MCA between rhinal fissure and olfactory fossa, and simultaneously tied left ICA permanently. We classified normal group and treated with Dangkijakyaksan group (sample group) and non treated group(control group). Medication was started on the 4th day after .occlusion, and stopped on ¢¥the 3rd month. On the 1st and 3rd month, we measured the concentrations of Estradiol(EO, LH, FSH with radioimunoassay method and killed rats, and fixed brains of them by intracardiac perfusion of 150cc normal saline and 10% formaldehyde solution, then we took out rat¢¥s brain, and sliced them per 2mm from frontal to caudate with brain slicer(Zivic miller Laboratories, PA., USA). We measured the left and right area of cerebral hemisphere by means of Image Analysis System. From this, we calculate the left and right ratio of area.
RESULTS:
After the occlusion of the left MCA, the infarcted area was formed in dorsolateral side of left hemisphere. The border between infarcted tissue and normal was well demarcated, and the brightness of infarcted tissue was slightly darker than that of normal. 2 months later, the cerebral atrophy progressed, and encapsulated scar was formed in infarcted hemisphere. 3 months later, the cerebral atrophy more progressed than that of 2 months, and transparent scar was formed in infarcted hemisphere, it¢¥s inner space contained liquid.
After 1 month of cerebral ischemic stroke, the concentration of E2 was 12.978.27pg/ml in normal group and 13.177.66pg/ml in the control group and 29.5212.90pg/ml in the sample group. The concentration of E2 in the sample group was significantly higher than in the others, but LH, FSH were insignificant. After 3 months of cerebral ischemic stroke, the concentrations of female sex hormones were insignificant difference among the groups.
When we measured the left and right ratio of cerebral hemisphere after 1 month of cerebral ischemic stroke, there wasn¢¥t significant difference between control and sample group. After 3 months of occulusion, there wasn¢¥t significant difference between control and sample group either.
CONCLUSION :
According to the result above, Dangkijakyaksan has a effect on preservation of estrogen and has no effect on cerebral atrophy.
