The Effect of Daejowhan(ÓÞðãü¯) on Cerebral and Thalamic atrophy following Middle Cerebral Artery Occlusion in Rats



Abstract
The purpose of this study is to find out the effections of Daejowhano Qla ,) on cerebral atrophy after middle cerebral artery occlusion in rats.
METHODS :
In this experiment, we used 18 Spraque-Dawley rats, weighing 280-340g. Cerebral infarction was induced from subtemporal method that is modified by Chen. Then we electrocauterized MCA between rhinal fissure and olfactory fossa, and simultaneously tied left ICA permanently. We classified treated with Daejowhan group(Sample Group) and non treated group(Control Group). Medication was started on the 4th day after occlusion, and stopped on the 3rd month. On-the 3rd month, we killed rats, and ¢¥fixed brains of them by intracardiac perfusion of 150cc normal saline and 10% formaldehyde solution, then we took out rat¢¥s brain, and sliced them per 2mm from frontal to caudate with brain slicer(Zivic miller .Laboratories, PA., USA). We measured the left and right area of cerebral hemisphere and thalamus by means of Image Analysis System. From this, we calculate the left and right ratio of area.
RESULTS :
After the occlusion of the left MCA, the infarcted area was formed in dorsolateral side of left: hemisphere. The border between infarcted tissue and normal was well demarcated, and the brightness of infarcted tissue was slightly darker than that of normal. 2months later, the cerebral atrophy progressed, and encapsulated scar was formed in infarcted hemisphere. 3months later, the cerebral atrophy more progressed than that of 2months, and transparent scar was formed in infarcted hemisphere, it¢¥s inner space contained liquid.
The left and right ratio of cerebral hemisphere was 0.640.14 in the Control Group, and 0.730.06 in the Sample Group on the slice level 2mm. On 4mm, Control Group 0.520.10, Sample Group 0.590.14. On 6mm, Control Group 0.590.15, Sample Group 0.560.09. On 8mm, Control Group 0.700.05, Sample Group 0.660.07. On 10mm, Control Group 0.760. 05, Sample Group 0.670.11.
The left and right ratio of thalamus in the section located 3.8mm posterior to the bregma was 0.640.02 in the Cotnrol Group, 0.620.07 in the Sample Group.
CONCLUSION :
On comparison of the left and right area of the cerebral hemisphere and thalamus, significant difference didn¢¥t exist between Sample Group and Control Group.
