Synthesis and breakdown of NAD^(+)-linked-¥á-glycerophosphate dehydrogenase in erythrocyte differentiation



Abstract
The synthesis and breakdown of cytosolic NAD^(+)-linked ¥á-glycerophosphate dehydrogenase (G3P; NAD^(+)-2-oxidoreductase, 1.1.1.8) were studied during induction of and recovery from phenylhydrazine-treated reticulocytosis in rabbit blood.
The enzyme activity was assayed according to the method of White and Kaplan.
The results obtained were asfollows:
1. Erythrocytes from normal rabbit contained about 2.5% of reticulocytes.
2. Activity of NAD^(+)-linked ¥á-glycerophosphate dehydrogenase was not detected in cytosolic fraction nomal rabbit erythrocyte.
3. The reticulocyte count in phenylhydrazine-treated rabbit to a value of about 55% on 5 day and obtained a value of 100% on 6 day.
4. Activity of cytosolic ¥á-glycerophosphate dehydrogenase of reticulocytes was maximal on 4 day with a value of 98 unit.
5. The activity of the cytosolic enzyme in initial stage after phenylhydrazine-treatment was inceased drastically (K_(S)=2.3814, Kd=0.00243, t1/2=28.52hrs.).
6. The activity constants following recovery from phenylhydrazine-treatment was; K_(S)=0.2288; Kd=0.0396, t1/2=18.78hrs.
7. An effort to prove the possible existane of glycerol phosphate shuttle during reticulocytes differentiation went for nothing the reason why the estimation of mitochondrial FAD^(+)-linked ¥á-glycerophosphate dehydrogenase activity by using Dawson and Thorne resulted in a failure.
